The incorporation of O6-methyldeoxyguanosine and O4-methyldeoxythymidine monophosphates into DNA by DNA polymerases I and

The modified nucleoside 5'-triphosphates 06-MedGTP and 04-MedTTP have been synthesised and their acceptability as DNA-precursors investigated using DNA polymerases I and
in an in vitro assay. 06-MedGMP is only incorporated into newly-synthesised DNA-like material in the presence of templates containing thymine bases. Similarly 04-MedTMP is only incorporated in the presence of templates containing guanine bases. The results confirm the promutagenic nature and base-pairing properties of 06-MeG and 04-MeT.
