RsrII-a novel restriction endonuclease with a heptanucleotide recognition site

A sequence-specific endonuclease present in extracts of Rhodopseudomonas sphaeroides 630 has been purified and characterized. The enzyme, Rsr II, recognises and cleaves the palindromic heptanucleotide sequence:

By virtue of its unusual specificity, RsrII cuts most DNA molecules very infrequently which should facilitate the physical mapping of large genomes.
