Sequence of the Dß2-Jß2 region of the human T-cell receptor ß-chain locus
Medical Research Council, Laboratory of Molecular Biology Hills Road, Cambridge CB2 2QH, UK
Received August 8, 1985. Accepted August 27, 1985.
We have sequenced the region encompassing a Dß2 segment and the Jß2 segments of the human T-cell antigen receptor ß-chain genes. The Dß2 element lies about 650 base pairs upstream of a cluster of seven potentially functional Jß2 sequences and one Jß2 pseudogene. Examination of human ß-chain cDNA sequences which involve rearranged Dß2 and Jß2 elements demonstrates that N-region, as well as junctional, diversity can occur during D-J Joining. Further, we present evidence for possible somatic mutation in active Jß2 segments.
